TGF-β and NF-κB signaling pathway crosstalk potentiates corneal epithelial senescence through an RNA stress response

class="figure-viewer-img"

Figure 3. TGF-β1 induces cellular senescence in HCECs. (A-B) The G1 phase arrest was induced by TGF-β1 treatment .Control and TGF-β1–treated HCECs were subjected to cell cycle analysis after 48h of culture. HCECs treated with H2O2 (200μM) were taken as positive control. A representative flow cytometric analysis of the DNA content was shown in (A) and the values are mean±SD (B). (C) HCECs were treated with TGF-β1 (10 ng/ml) alone, or in combination with LY364947 (2μM) for 3days, and tested for SA-β-Gal activity. (D) The percentage of SA-β-gal -positive cells in HCECs. (E-F) The mRNA (E) and protein (F) expression of p16 and p21 in HCECs induced by TGF-β1. Bar graphs represent mean±SD. **P≤0.01,*P ≤0.05 vs. control. Data are representative of three independent experiments.
