Figure 2. Effect of miR-214-3p on proliferation, apoptosis and inflammatory response on chondrocytes. (A) CCK-8 analysis showing effect of miR-214-3p mimic and inhibitor on cell proliferation for 24 and 48 hours. (B) Colony formation assay on chondrocytes exposed to miR-214-3p-mimic or inhibitor. (C) Flow cytometry analysis for studying apoptosis index after exposure to miR-214-3p mimic or inhibitor for 24 hours. (D) Cell cycle progression analysis evaluating the effect of miR-214-3p mimic or inhibitor treatment for 24 hours. (E) qRT-PCR analysis for relative mRNA levels of miR-214-3p, IL-1β, IL-6 and TNF-α after exposure to miR-214-3p mimic or inhibitor for 24 hours. (F) Western blot analysis for expression of inflammatory proteins IL-1β, IL-6 and TNF-α after exposing the chondrocytes to miR-214-3p mimic or inhibitor. The results are presented as mean (n=3) ± %RSD. *P<0.05, **P<0.01, ***P<0.001 compared to normal control.
