IL-6, p-SAPK/JNK and SAPK/JNK antibodies were used. (C) Cultured PC3 cells were treated with C10 in the presence of different inhibitors (siPKCδ and the JNK-specific inhibitor Tanzisertib [CC-930]) for 24 h. The cells were then stained with Hoechst 33258 and photographed using a fluorescence microscope (magnification ×200, scale bar: 100 μm). (D, E) Cultured PC3 cells were stained with annexin-V-FITC and PI for flow cytometry analysis. (F) The mRNA levels of PKCδ, Caspase-9, IL-6, IL-8, IL-1β and Bax were measured by qRT-PCR.
