Figure 1. Construction of DDP-resistant GC cell line (SGC7901/DDP). (A) Cell viability assay demonstrated higher survival rate of SGC7901/DDP compared to normal SGC7901 cell line upon treatment with DDP. (B) BrdU assay showed that the DDP-resistant SGC7901 cell line had significantly higher survival rate compared to parental SGC7901. (C) Results from flow cytometry showed that the apoptotic rate of SGC7901/DDP cells was significantly lower compared to parental SGC7901. (DE) Result of the transwell chamber assay indicated increased migration and invasion in SGC7901/DDP. (F) Result from the wound healing assay further verified the stronger invasive ability of SGC7901/DDP. (G) Result from western blot demonstrated lower apoptotic rate, stronger invasion and migration, increased drug resistance induced by higher expression of Bcl-2 family proteins, activation of EMT and overexpression of MDR1 and ABCC1 proteins, respectively. ***p<0.001, **p<0.01 compared to parental SGC7901.
