Figure 2. Effects of CAMKIIγ on proliferation and cycle progression of MM cells. (A) CAMKIIγ and Stat3 protein expression in human MM cell lines, including ARP-1, MM1.S, 8226, KM3, U266 cell lines. (B) CAMKIIγ overexpression or knockout efficiency was measured by Western blot. GAPDH was used as a loading control. (C) Proliferation curves of KM3 or U266 stable-transfected cells with CAMKIIγ overexpression or knockout, compared with their controls. The MTT assays were performed for a total of a week (*P < 0.05, **P < 0.01). (D) KM3 cells of CAMKIIγ overexpression and the control were cultured in serum-free medium for 48 hours, then maintained in 1640 medium supplemented with 15% fetal bovine serum at the indicated times. Representative images and quantification of cell cycle by flow cytometry (*P < 0.05, ***P < 0.001). (E) Representative images and quantification of cell cycle in U266 cells after DOX-induced CAMKIIγ-KO (***P < 0.001).
