CLCA4 inhibits cell proliferation and invasion of hepatocellular carcinoma by suppressing epithelial-mesenchymal transition via PI3K/AKT signaling

Figure 5. CLCA4 expression was correlated with the expression of epithelial-mesenchymal transition (EMT) markers. (A) Serial sections of human HCC tissue were subjected to IHC staining with antibodies against CLCA4, E-cadherin, Vimentin and N-cadherin. In patient #1, low CLCA4 expression in HCC tissues was accompanied by the absence of E-cadherin and elevated Vimentin, N-cadherin. In patient #2, high CLCA4 expression was accompanied by elevated E-cadherin and the absence of Vimentin, N-cadherin. The scale bar represents 50 μm. (B) CLCA4 expression was positively associated with E-cadherin expression and negatively correlated with vimentin and N-cadherin expression. (C) Decreased expression of E-cadherin with increased expression of Vimentin and N-cadherin in CLCA4-silenced cells compared with the control cells. An opposite expression pattern of these genes was observed in CLCA4-transfected cells. (D) The mRNA of E-cadherin was down-regulated, while the vimentin and N-cadherin was up-regulated, when CLCA4 was silenced. And opposite expression was observed in CLCA4-transfected cells.
